چکیده

Abstract Myostatin, a member of the transforming growth factor-? superfamily, is an attractive target for muscle disease therapy because its role as negative regulator and strength. Here, we describe novel antibody therapeutic approach that maximizes potential myostatin-targeted therapy. We generated antibody, GYM329, specifically binds latent form myostatin inhibits activation. Additionally, via “sweeping technology”, GYM329 reduces or “sweeps” in plasma. Compared with conventional anti-myostatin agents, surrogate exhibit superior strength-improvement effects three different mouse models. also demonstrate efficacy due to specificity sweeping capability. Furthermore, show increases mass normal cynomolgus monkeys without any obvious toxicity. Our findings indicate improve strength patients muscular disorders.

Anti-myostatin antibody increases muscle mass and strength and improves insulin sensitivity in old mice.

Sarcopenia, or skeletal muscle atrophy, is a debilitating comorbidity of many physiological and pathophysiological processes, including normal aging. There are no approved therapies for sarcopenia, but the antihypertrophic myokine myostatin is a potential therapeutic target. Here, we show that treatment of young and old mice with an anti-myostatin antibody (ATA 842) for 4 wk increased muscle ma...

Antibody-directed myostatin inhibition enhances muscle mass and function in tumor-bearing mice.

Cancer cachexia describes the progressive skeletal muscle wasting and weakness in many cancer patients and accounts for >20% of cancer-related deaths. We tested the hypothesis that antibody-directed myostatin inhibition would attenuate the atrophy and loss of function in muscles of tumor-bearing mice. Twelve-week-old C57BL/6 mice received a subcutaneous injection of saline (control) or Lewis lu...

Myostatin Neutralization Results in Preservation of Muscle Mass and Strength in Preclinical Models of Tumor-Induced Muscle Wasting.

Skeletal muscle wasting occurs in a great majority of cancer patients with advanced disease and is associated with a poor prognosis and decreased survival. Myostatin functions as a negative regulator of skeletal muscle mass and has recently become a therapeutic target for reducing the loss of skeletal muscle and strength associated with clinical myopathies. We generated neutralizing antibodies ...

Lower skeletal muscle mass in male transgenic mice with muscle-specific overexpression of myostatin.

Mutations in the myostatin gene are associated with hypermuscularity, suggesting that myostatin inhibits skeletal muscle growth. We postulated that increased tissue-specific expression of myostatin protein in skeletal muscle would induce muscle loss. To investigate this hypothesis, we generated transgenic mice that overexpress myostatin protein selectively in the skeletal muscle, with or withou...

Modulating skeletal muscle mass by postnatal, muscle-specific inactivation of the myostatin gene.

By using a conditional gene targeting approach exploiting the cre-lox system, we show that postnatal inactivation of the myostatin gene in striated muscle is sufficient to cause a generalized muscular hypertrophy of the same magnitude as that observed for constitutive myostatin knockout mice. This formally demonstrates that striated muscle is the production site of functional myostatin and that...
